That bs mote is not on the camera sensor, and appears on any filters, and no way I can calibrate it out… pure drama :-)
This should actually be manageable.
How have you calibrated your flats? How does the integrated master flat look like.
Do you have the opportunity to create new flats? It looks like your skyflats have a heavy gradient.

Are you using BIAS or Darkflats to calibrate?
Have you blinked through all Flats / Darkflats / Bias to see if there are any outliers?
Have you calibrated your lights properly with matching Darks?
